In Strike the Blood ist der vierte Primogenitor der stärkste Vampir der Welt, der eigentlich nur in Legenden existiert. Begleitet von zwölf "Kenjuu" und Katastrophen bringend, erscheint dieser Vampir in Japan. Um diesen vierten Primogenitors zu überwachen und zu vernichten, entscheiden sich die Regierung und der "Lion King" einen Angriffs-Magier, bekannt als "Schwert-Schamane", zu entsenden. Jedoch ist diese Schwert-Schamanin aus irgendeinem Grund ein junger Lehrling namens Himegari Yukina. Mit dem stärksten anti-primogenitor Geisterspeer ausgerüstet, trifft Yukina im Dämonen-Bezirk, Itogami City, ein. Was ist die wahre Identiät des vierten Primogenitors, Akatsuki Kojou, dem sie dort begegnet?!
